<P>PROBLEM TO BE SOLVED: To provide a method for readily and efficiently extracting and purifying a nucleic acid from a biological material having a hard cell wall such as a bacterium of the genus Mycobacterium present in a biological sample such as excrement from which the purification is considered to be difficult. <P>SOLUTION: The method for simultaneously extracting and purifying the nucleic acid comprises stirring the sample such as the excrement containing the biological material such as the one belonging to the Mycobacterium, with an aqueous solution containing a chelating reagent and a quaternary ammonium salt, an organic solvent and fine particles. For example, the chelating agent is selected from EDTA or a reagent resembling the EDTA.
