Proteolytic enzyme inhibitors are purified (concentrated) by contacting an impure aqueous solution of the inhibitor with a waterinsoluble polymer-enzyme combination material whereby at least some of the inhibitor is sorbed by the combination material, separating the combination material from the unsorbed aqueous phase, and desorbing a purified aqueous solution of the inactvator from the combination material, the latter having been formed by contact of a proteolytic enzyme, inhibitable by the inhibitor, with a polymeric material cross-linkable by this proteolytic enzyme. The inhibitor may be trypsin, kunitz or kallikrein inhibitors; the enzyme may be trypsin, chymotrypsin or kall-krein and examples of suitable polymeric material are ethylene-maleic anhydride copolymer, maleic anhydride-vinylpyrrolidone copolymer and methacrylic acid-methacrylic acid-3-fluoroanilide copolymer. In a modification of the process, the combination material is water-soluble and, as the impure aqueous solution, serum containing the inactivator is used. The inhibitor is subsequently desorbed by shifting the pH and/ or modifying the ion concentration and/or displacement by means of competitive inhibitors or enzymes e.g. tryptamine, n-butyl-amine or epsilon-amino caproic acid for trypsin, and/ or by the addition of substances, such as urea, which are capable of loosening or dissolving molecular interactions, possibly by a reversible denaturation of the proteins concerned.
