<P>PROBLEM TO BE SOLVED: To elucidate substitution of an amino acid bringing about lowering of transcription-activating ability to an androgen receptor (AR) and develop a method for examining existence of such substitution of the amino acid. <P>SOLUTION: In an artificial animal cell or the like, a variant AR containing an amino acid having the following properties (a) and (b) is expressed and a reporter gene in which a base sequence encoding a reporter protein is connected to the downstream of an androgen-responsive sequence and a base sequence necessary for initiation of transcription is introduced into its chromosome: (a) The amino acid is located at one or more positions selected from positions corresponding to specific amino acids of an amino acid sequence in alignment based on a homology of an amino acid sequence and is different from an amino acid located at the position of the amino acid sequence of a standard AR. (b) A property in which an ability to activate transcription of the reporter gene contained in a chromosome of an animal cell in the presence of androgen is lower than that of a standard AR is imparted to the variant AR having the amino acid. <P>COPYRIGHT: (C)2004,JPO |
