摘要 <P>PROBLEM TO BE SOLVED: To provide an improved method for attaining a single base resolution of fragments for DNA sequence determination by giving a low viscosity medium easily chargeable/dischargeable into/from a capillary electrophoretic tube, and also to provide a device therefor. <P>SOLUTION: This method for determining the nucleotide sequence of a target polynucleotide comprises separating DNA fragments in a mixture for DNA sequence determination based on their difference in dimension. Specifically, the method includes the step of separating the DNA fragments by capillary electrophoresis in a modified aqueous solution containing a medium for such electrophoresis. Thereby, a single base resolution is attained for the fragments each with at least about 300-base length. Besides, the low viscosity of the solution allows quick charging and recharging of such solution into the capillary tube. <P>COPYRIGHT: (C)2004,JPO
