1,161,108. Tricyclo[3.2.2.0<SP>2'4</SP>]nonane derivatives; penicillin salts. E. I. DU PONT DE NEMOURS & CO. 14 March, 1967 [29 March, 1966], No. 11949/67. Headings C2A and C2C. Novel compounds of formula wherein X and Y are each H, CH 3 or C 2 H 5 , R is H or C 1-4 alkyl, R 1 and R 2 are each H, C 1-4 alkyl or allyl and n is 0 or 1 and acid addition salts thereof including the salt with penicillin, are prepared by formation of an amine group by action of O - methylhydroxylamine on the required nonane - 1 - methyl magnesium halide, by hydrolysis of the corresponding carbamide, or by reduction of the corresponding amide or oxime, optionally followed by introduction of R 1 and/or R 2 where at least one is not H, and/or salt formation. Tricyclo[3.2.2.O<SP>2</SP>'<SP>4</SP>]nonane - 1 - - carboxylic acid esters are prepared by action of CH 2 I 2 and a Zn/Cr couple on the corresponding bicyclo[2.2.2]oct - 2 - ene - 1 - carboxylic acid esters and then hydrolysed to the acids which may be converted via the isocyanate into the carbamide, or into the acid chloride which on reaction with ammonia gives the amide or on reaction with cadmium or lithium alkyls gives the 1-acyl compound which is then converted to its oxime. Halides (halogen for NR 1 R 2 and n = 1 in the above formula) are prepared by action of thionyl chloride on the corresponding alcohols which are prepared by reaction of Grignard reagents with either the required 1-carboxylic acid chlorides or 1-acyl compounds. Pharmaceutical compositions having anti-viral activity and being in conventional forms for oral or parenteral administration comprise an above novel compound or salt thereof and a carrier or diluent therefor.
