High-throughput screening of gene function using libraries of plasmids in a multi-well format for functional genomics applications

A library of a multitude of unique expressible nucleic acids comprises a multiplicity of compartments. Each of the compartments comprises one or more adenoviral vectors comprising at lest one unique expressible nucleic acid of the library in an aqueous medium. The adenoviral vector is capable of introducing the unique expressible nucleic acid into a host cell, capable of expressing the product of the unique expressible nucleic acid into the host cell and is deleted in a portion of the adenoviral genome necessary for replication thereof in the host cell. Also described is a method for producing a library comprising of a multitude of unique expressible nucleic acids arranged in a multiplicity of compartments, a method of producing a recombinant adenovirus vector library and a method for measuring the function of a unique expressible nucleic acid(s) of unknown function in a high throughput setting.
