<p>A general process for the identification of trace levels of tagged proteins present in complex biological systems is described. The process involves the enzymatic or chemical digestion of the biological system, the selective retrieval of tagged peptides from the digested system using an affinity recognition system, mass spectrometry of the affinity-isolated species and protein identification via database search using the masses of the retrieved species and the specificity of the enzyme/chemical. Methods for sample preparation, database search and data validation are given.</p> |
