L-Lysine is prepared by culturing a threonine- or methionine-sensitive strain of the genus Brevibacterium on an aqueous nutrient medium under aerobic conditions, maintaining the pH of the medium between 5 and 9, and recovering the L-lysine. Specified micro-organisms are strains of B. flavum (A.T.C.C. Nos. 21127, 21128 and 21129) obtained by mutation with X-rays,γ-rays, U.V. light, or by treatment with sodium nitrite solution, ethyl methane sulphonate, or N - methyl - N<SP>1</SP> - nitro - N - nitrosoquaniline. The threonine-senstive strains require threonine for growth but are inhibited by concentrations of threonine in excess of 10 mg./mol., the preferred concentration being between 0À01 mg. and 1 mg./mol. medium. Suitable carbon sources include carbohydrates, particularly glucose, and nitrogen is preferably provided by ammonium salts of organic or inorganic acids.
