<P>PROBLEM TO BE SOLVED: To provide a method for readily obtaining a cytokine agonist peptide by using an Escherichia coli random peptide library. <P>SOLUTION: The Escherichia coli random peptide library in which a supporting protein containing a cysteine loop is expressed on the surface and a random peptide is expressed in the cysteine loop is used as the library. Two- stage screening is carried out to the library. In the primary screening stage, the library is bound to a receptor of a substance which is well-known as an agonist of the cytokine, and selected by panning. In the secondary screening, a replicated product of a clone selected by the primary screening is reacted with a host in which the cytokine promoter and a reporter gene are introduced and expression of a reporter protein is observed. As a result, it is detected that the peptide is the agonist peptide transmitting a signal to the cytokine. <P>COPYRIGHT: (C)2004,JPO |
