New hybrid DNA, useful for measuring non-homologous recombination processes and for identifying modulators of these processes, which are potentially useful as anticancer and antiretroviral agents

An isolated double-stranded hybrid DNA (I), intended as substrate for non-homologous recombination, comprising: (a) a single unidirectional promoter and at least two reporter genes (A and C) in the direction of transcription, with only (A) being controlled by the promoter; and (b) two identical sites for targeted cleavage, one between the promoter and (A), the other upstream of, or within, (C), is new. Independent claims are also included for: (1) a vector that contains (I); (2) a cell modified by (I) or the vector of (1); (3) a recombinant non-human mammal that contains (I); (4) measuring non-homologous recombination processes (non-homologous end-joining (NHEJ)) in mammalian cells; (5) screening for modulators of NHEJ in mammalian cells; and (6) a kit for use in the method of (4).
