PROBLEM TO BE SOLVED: To provide a method of manufacturing a filler for liquid chromatography which enables good optical isomer separation of a target compound. SOLUTION: In the method of manufacturing a filler for liquid chromatography, a process where the required amount of a dope of a polysaccharide derivative is divided into a plurality of parts and a part of the dope of the polysaccharide derivative is applied to a porous carrier to be dried is repeated a plurality of times, and the required amount of the polysaccharide derivative is carried by the porous carrier. The quantity of the carried polysaccharide derivative in the filler is 20 mass% or higher. A TS (tetrakis(trimethylsilyl)silane) coefficient obtained by using a column for optical isomer separation for liquid chromatography prepared by packing the filler into a column pipe is in the range of 0.25 to 1.0.
