PURPOSE: A DNA replicon of hepatitis C virus(HCV), a transformed cell line containing the same and a method for detecting HCV using the same cell line are provided. The infection of HCV is accurately detected because all of the proteins of HCV expressed in the higher cells are used. CONSTITUTION: DNA replicon of HCV comprises internal ribosomal entry site(IRES) of HCV, IRES of poliovirus, a selection marker and encephalomyocarditis(EMCV), wherein the non-structural gene region from NS3 to NS5B of HCV, linked to the 3'-terminal of EMCV IRES and poly A tail are contained in the replicon; one or more kinds of polynucleotides selected from the core gene, E1 gene, E2 gene, p7 gene and NS2 gene of HCV are inserted between the HCV IRES and poliovirus IRES; and the replicon is a plasmid(KCTC 10226BP). A RNA transcript derived from the DNA replicon of HCV is provided. A transformed cell line is produced by transforming a cell line with the RNA transcript, wherein the cell line is human liver cancer cell line(KCTC 10226BP). A method for detecting HCV comprises independently reacting an extract of the cell line transformed with the RNA transcript of HCV and an extract of the untransformed cell line with samples and comparing the reaction results, wherein the reaction results are analyzed by using western blotting or ELISA.
