<p>Materials and methods useful in the in situ generation of viral vectors are described. Briefly, virus is liberated from cells by treatment with polysorbate and tri-butyl phosphate yielding non-viscous lysates with relatively little cell debris. Lysis is followed by step density ultracentrifugation to isolate crude virus. Finally, purification and helper removal is accomplished by dynamic banding ultracentrifugation.</p> |
