<p>The present invention provides a method of producing soluble, correctly folded, and functional proteins, especially proteins of large sizes (> -400 amino acids) in large quantity (> milligrams) in E. coli or other host cells. The method involves splitting the large proteins into smaller pieces and co-expressing them in the same cell. Proper split sites can be determined by three dimensional structure analysis, biochemical and/or sequence analyses.</p> |
