PROBLEM TO BE SOLVED: To provide a transmission assembling structure of a motorcycle, increased in assembling workability and rigidity of the engine by modulating the engine and the transmission. SOLUTION: In the engine 3 of the motorcycle 1 comprising the transmission 60, a crankshaft 9, an input shaft 37, a clutch shaft 38 and an output shaft 39 are arranged in parallel inside an engine case 34 and constituted separably approximately in the vertical direction for the shaft center direction. The transmission 60 comprises a transmission case 61 constituted as a different body. A drive shaft 62 and a driven shaft 63 are arranged in parallel inside the transmission case 61. One end parts of the drive shaft 62 and the driven shaft 63 respectively are protruded outside the transmission case 61. The transmission case 61 is constituted separably approximately in the vertical direction for the shaft center direction and fastened to the vehicular left side engine case 36b by a fastening bolt 64b via the vehicular right side engine case 36a.
