<p>The invention relates to a non-viral method of transfecting cell cultures, tissue and/or organ cultures and can also be used for in vivo gene transfer when observing particular basic conditions. The invention is suitable for medical and biological basic research, particularly regarding preliminary studies on gene therapy and in the pharmaceutical industry. The method for gene transfer into animal cells is characterized in that vector DNA in aqueous solution is packed into compact nanoparticles by successive addition of oppositely charged polyelectrolytes and that transfection of the cells is performed thereafter. Furthermore, the method is characterized in that the vector DNA - is made to interact with polycations and condensed in a first step, - is mixed with polyanions to coat a layer in a second step, and that these steps are repeated, if necessary. For in vivo gene transfer, the vector DNA is packed into polyelectrolyte nanoparticles (shells) which are mixed with e.g. Pluronic F127 gel and, following exposure of the tissues/organs/tumors to be transfected, applied thereon in liquid form.</p> |
