METHOD OF CULTURING RECOMBINANT YEAST FOR ENHANCING PRODUCTIVITY OF RECOMBINANT PROTEIN

PURPOSE: A method of culturing recombinant yeast for enhancing the productivity of recombinant proteins is provided, thereby mass-producing recombinant proteins under regulation of GAPDH(alyceraldehyde-3-phosphate dehydrogenase) promoter. CONSTITUTION: A method of culturing recombinant yeast for enhancing the productivity of recombinant proteins comprises the steps of: (i) feeding an additional medium to the culture medium in a constant rate after 20 to 28 hours of seed inoculation; and (ii) linear feeding the additional medium to the culture medium to maintain the specific growth rate of the recombinant yeast to 0.05 to 0.08 h-1, wherein the feeding rate of the additional medium in the step (i) is 0.8 to 1.3 g/L/hr; the concentration of the recombinant yeast in the step (i) is 10 to 15 g/L; the additional medium feeding in the step (ii) is carried out until the concentration of the recombinant yeast is 60 g/L; and the linear feeding is calculated by the following equations, F=M(t-t0)+B, M=2Δ V/t12-t02, and dF=μ*d(XV)0/S0Yx/s, wherein F is medium feeding rate(ml/hr), M is proportional factor(ml/hr2), B is feeding correction value(ml/hr), ΔV is total volume of the additional medium(ml), t1 is termination time of the medium addition(hr), t is fermentation time(hr), t0 is initial medium linear feeding time(hr), dF is change of medium feeding rate(ml/hr), μ* is determined specific growth rate of cell, X is cell concentration(g/L), V is volume of culture medium(ml), S0 is initial medium concentration(g/L), and Yx/s is yield of cell to substrate.
