PURPOSE:To improve the hydrolytic reaction rate than that in using alginate lyase alone, by using an alkaline earth metal and/or surfactant in hydrolyzing alginic acid or salts thereof with the above-mentioned enzyme. CONSTITUTION:A strain, belonging to the genus Xanthomonas and having the ability to produce alginate lyase, e.g. Xanthomonas sp. N-26 (FERM-P No. 8800), is cultivated in a culture medium prepared by adding preferably 0.1-1% alginic acid and salt thereof to meat extract, peptone, yeast extract, malt extract, corn steep liquor, casamio acid, etc., preferably at 27-36 deg.C for 5-40 hr to give alginate lyase from the cultivated microbial cells. In the process, the above-mentioned enzyme is used in an amount of about 1 unit/ml. Furthermore, a substance having the chelating ability with an alkaline earth metal, e.g. citrate, etc., is used in a mol concentration of >=1.6 times based on the amount of the metal alginate to be hydrolyzed and a surfactant in an amount of preferably 0.05-1% (wt.vol. is added to enhance the hydrolytic ability of the alginate. |
