<P>PROBLEM TO BE SOLVED: To provide a new alkaline protease having strong activity on insoluble proteins such as keratin as well as soluble proteins. <P>SOLUTION: The alkaline protease has the following physical and chemical properties. (a) Action and substrate specificity: it has an action on proteins and peptides to break the peptide bond by an endo-type mechanism and produce low-molecular oligopeptides and amino acids and further exhibits strong action on insoluble proteins such as keratin, (b) stable pH: it is stable at pH 1.5-12.0 under the treating condition of 30°C and 24 hr, (c) optimum pH: the optimum pH is 11.0-11.5 when casein is used as the substrate and ≥12.0 when keratin is used as the substrate, and (d) specific activity: the specific activity is about 1,100 (PU/mg protein) when casein is used as the substrate and about 3,300 (KU/mg protein) when keratin is used as the substrate. <P>COPYRIGHT: (C)2004,JPO |
