DNA POLYMERASE COMPOSITION FOR NUCLEIC ACID AMPLIFICATION

PROBLEM TO BE SOLVED: To provide a heat-resistant DNA polymerase composition having excellent amplification efficiency of nucleic acid.

SOLUTION: The DNA polymerase composition for the amplification of nucleic acids contains a modified heat-resistant DNA polymerase (the 1st DNA polymerase) having a 3'-5' exonuclease activity corresponding to 0-5% of the activity before modification and a heat-resistant DNA polymerase having a 3'-5' exonuclease activity or another modified heat-resistant DNA polymerase (the 2nd DNA polymerase) having a 3'-5' exonuclease activity corresponding to 100-6% of the activity before modification. The 1st and 2nd DNA polymerases have a DNA synthesis rate of ≥30 bases/sec and a thermal stability to hold ≥60% residual activity after the treatment at pH 8.8 (measured at 25°C) at 95°C for 6 hr.
