<p>A method of analyzing a peptide for determining C-terminal amino acid sequence based on successive dissociation reactions of C-terminal amino acids, in which C-terminal amino acids of a peptide can be successively dissociated while inhibiting undesirable side reactions such as the cleavage of a peptide linkage present within the peptide and the chemical treatment can be performed under general conditions. In the method, an alkanoic anhydride and a perfluoroalkanoic acid both in a vapor state supplied from a mixture obtained by adding a small amount of the perfluoroalkanoic acid to the alkanoic anhydride are caused to act on a dry peptide sample in a dry atmosphere at a temperature selected from the range of 15 to 60°C to dissociate a C-terminal amino acid through the formation of a 5-oxazolone structure and cleavage of the 5-oxazolone ring. The C-terminal amino acid sequence is specified based on the decreases in molecular weight of the reaction products thus obtained in series.</p> |
