<p>PURPOSE: A PNA(peptide nucleic acid) monomer having photo-labile protecting group is provided, which PNA monomer is used in production of PNA oligomer, PNA/DNA chimera and PNA/peptide conjugate, especially PNA chip. CONSTITUTION: A PNA monomer represented by formula (1) is provided, wherein R is a compound of R1, R2, R3, R4 and R5; R1, R2, R3 and R4 are the same or different, and independently hydrogen, nitro, nitrile, halogen atom, (C1-C4) alkyl, (C1-C4) alkoxy, acyl or aryl, or R2 and R3 forms 5 to 6 membered ring containing two oxygens; R5 is H, (C1-C4) alkyl or aryl; n is 0 or 1; and B is protected or unprotected, natural or synthetic nucleic acid base. A method for preparing the compound of formula (1) comprises reacting a compound of formula (5) with a compound of formula (6), wherein the compound of formula (5) is prepared by (a) reacting a compound of formula (6) with a compound of formula (2) to prepare a compound of formula (3), (b) hydrolysis of the compound of formula (3) and reacting it with di-t-butyl dicarbonate to prepare a compound of formula (4), and (c) cyclization of the compound of formula (4) and deprotecting the compound.</p> |
