Retroviral particles with deletions in E1 and E4 regions characterised by the sequence of interest being present in between two long repeat regions (LTR)

A composition comprises a retroviral vector and genetic elements encoding for retroviral proteins that complement the retroviral vector to allow for the constitution of a retroviral particle. The vector and genetic elements being incorporated into one or several recombinant adenoviruses defective for all or part of at least the E1 and E4 regions. Preferably the genetic elements comprise a nucleic acid coding for a retroviral gag protein, a nucleic acid coding for a retroviral pol protein and a nucleic acid coding for an envelope protein and the vector comprises a nucleic acid comprising, between two LTR regions, a retroviral packaging sequence and a nucleic acid sequence of interest. The compositions can be used for the preparation of a product for transferring nucleic acid into cells
