Methods are provided for regulating G protein mediated signal transduction in a cell that exhibits aberrant G protein mediated signal transduction by expressing a non<endogenous G protein coupled receptor (GPCR), which can be a constitutively activated non?endogenous GPCR, which can associate with an endogenous G protein in the cell, or a chimeric non?endogenous GPCR, comprising a first GPCR, which can be an orphan GPCR, modified to contain an operatively linked heterologous ligand binding domain and which, when contacted with an endogenous ligand or a non?endogenous ligand, can associate with an endogenous G protein in the cell. Also provided are methods of ameliorating a pathologic condition associated with aberrant GPCR signal transduction in cells of a subject having or predisposed to the pathologic condition, for example, a myopathy or a neuropathy, by administering a polynucleotide encoding a constitutively activated non?endogenous GPCR to the subject. |
