It is intended to disclose a reagent for measuring alanine aminotransferase activity which contains L-alanine, 2-oxoglutaric acid, lactate dehydrogenase and reduced nicotinamide adenine dinucleotide, characterized by further containing a substance having an effect of inhibiting lactic acid dehydrogenase activity. It is also intended to disclose a method of measuring alanine aminotransferase activity characterized by comprising bringing a sample likely containing alanine aminotransferase into contact with L-alanine, 2-oxoglutaric acid, lactate dehydrogenase, reduced nicotinamide adenine dinucleotide and a substance having an effect of inhibiting lactic acid dehydrogenase activity. According to the above reagent or the above measurement method, an increase in the reagent blank (i.e., an increase in the initial absorbance) can be suppressed.
