PURPOSE: Method for freeze-preserving a zygote with a plastic straw is provided, which increase the survival rate of zygotes, and prevents them from being contaminated when thawing after freeze-preserving. CONSTITUTION: Method for preparing a plastic straw for freeze-preserving a zygote comprises the following steps: (i) A first step to collect zygotes, and cultivate in mHTF added of 10% FCS until the blastocyst stage; (ii) A second step to freeze-preserve the zygotes in the blastocyst stage with freeze-preserving solution EFS or EDS by using a plastic straw of 0.2-0.5mm diameter which is sterilized in a flowing 70% ethanol and dried in a clean bench; and (iii) A third step to research the survival rate by exposing zygotes to VS1 which is prepared by adding 7.5% ethylene glycol and 7.5% DMSO to a basic solution(TCM-199 + 20% FCS). As the result of research, the method of using a plastic straw can be used to both of EFS and EDS without significant differences. The survival rate of zygotes in early stage is less than in 2-, 4-, 8-cell stage, morula, the blastocyst stage.
