<P>PROBLEM TO BE SOLVED: To provide a plasmid vector capable of artificially controlling phenanthrene-decomposing function of a microorganism of Sphingomonas xenophaga AJ1 strain by elucidating the information transmission mechanism of the microorganism. <P>SOLUTION: A plasmid including a group of enzyme genes relating to the decomposition of phenanthrene is found from the Sphingomonas xenophaga AJ1 strain. The plasmid vector is a plasmid vector including a replication protein gene derived from pSXAc2 and an autonomous replication sequence and includes five phenanthrene decomposition enzyme genes, genes encoding a protein having unclear functions and a replication protein, and an autonomous replication sequence. A decomposing ability to phenanthrene or the decomposition or metabolic products can be imparted to the Sphingomonas by introducing the plasmid into the microorganism. The decomposing ability to phenanthrene is artificially controlled by elucidating the information transmission mechanism of the AJ1 strain using a replication protein and an autonomous replication sequence formed by deletion, addition or substitution of an amino acid or a nucleotide. <P>COPYRIGHT: (C)2003,JPO |
