摘要 |
PROBLEM TO BE SOLVED: To provide a microscope which can detect the light emitted from a micro-region with high efficiency even a as small as possible photoirradiation and can rapidly measure a sample. SOLUTION: The sample 18 dyed with molecules having three electronic states including at least a ground state is irradiated with a first lightλ1 causing the transition of the molecules from the ground state to the first excitation state and a second lightλ2 causing the transition of the molecules from the first excitation state to the second excitation state of a higher energy level through an optical system having an objective lens 16 and a solid immersion lens 17 so that at least portions thereof overlap each other and the light emitted from the sample is detected, thereby, the light emitted from the micro-region is detected with the high efficiency with a as small as possible photoirradiation and the microscopic image of the excellent super resolution and detection sensitivity capable of rapidly measuring the sample is obtained.
