A method for detecting mutations in a nucleic acid based on the application of a tensile force to separate hybridized oligonucleotides, is new. Detecting mutations in a nucleic acid (NA) comprises: (a) treating many NA strands with at least two type of oligonucleotides, receptor oligos and probe oligos, so as to form, by hybridization, sample complexes (SC) and reference complexes (RC) such that an immobilized, or immobilizable, linkage (L) between SC and RC is formed; (b) optionally immobilizing (L); (c) applying a tractive force to (L); and (d) determining which of the complexes has separated.
