PIG RAG-1 GENE AND USE THEREOF

PROBLEM TO BE SOLVED: To provide a DNA which codes a pig DNA protein, and to provide a method for producing a pig in which a pig RAG-1 gene function is repressed.

SOLUTION: In this method for producing the pig, a BAC clone containing the pig RAG-1 gene is obtained by screening pig genome DNA libraries. Three clones are obtained from the BAC clone by sub-cloning a region containing the RAG-1 gene. Base sequences are determined by sequencing the clones and then mapping is conducted by restriction enzyme digestion, so that a gene map is formed. A targeting vector which knocks out the RAG-1 gene is formed based on information of the gene map. The endogeneous pig RAG-1 gene function is repressed by utilizing the vector, so that the pig which has no acquired immunity is produced.
