PURPOSE: A process for preparing water soluble beta-glucan having physiological activity with a high yield from an easily obtainable and cheap starting material, i.e., flammulina velutipes fruit body is provided. CONSTITUTION: The process for preparing water soluble beta-glucan from flammulina velutipes fruit body comprises the steps of adding the fruit body as it is, or in a cut or ground form to water and heating them to extract water soluble beta-glucan. Also, the process further comprises the step of adding an organic solvent such as 1,3-butylene glycol and propylene glycol. In another variant, the process comprises adding a small amount of water to flammulina velutipes fruit body as it is, or in a cut or ground form, maintaining at 70 to 90 deg.C for about 4 hours to deactivate beta-glucanase, treating with an enzyme to decompose saccharide, protein, carbohydrate and cellulose, maintaining at 30 to 45 deg.C for about 60 minutes, adding a predetermined amount of water and then heating them to extract beta-glucan.
