PROBLEM TO BE SOLVED: To provide high detection accuracy by enabling the amplification of SPR response without damaging the original advantage of SPR spectroscopy and provide a method for detecting a biochemical specimen and a detecting chip for SPR spectroscopy comprised of simple processes and capable of simplifying a process for detecting the biochemical specimen with satisfactory reproducibility without requiring excess workmanship of a measurer. SOLUTION: A gold membrane is formed on a surface of a glass board. By such a constitution as to form a loop structure of each probe DNA and direct a free terminal side to the side of the membrane when the probe DNA is arranged on the membrane, hybridization is brought about in the case of the presence of a target gene. By the resolution of the loop structure, only hybridized DNA probes are selectively modified by colloidal gold. Therefore, it is possible to obtain a large difference in a SPR angle shift in which a shift due to the colloidal gold modification is added to a hybridization shift in SPR measurement.
