Packaging cell lines for use in facilitating the development of high capacity adenoviral vectors

A packaging cell line expressing an adenovirus structural protein, polypeptide or fragment thereof, selected from a penton base, a hexon, a fiber, a polypeptide IIIa, a polypeptide V, a polypeptide VI, a polypeptide VII, a polypeptide VIII, biologically active fragments thereof, wherein the gene encoding said protein, polypeptide, fragment comprises a tripartite leader sequence. Also disclosed are viral vectors and compositions of viral vectors, wherein said vectors comprise deletions or mutations of a DNA sequence encoding an adenovirus fiber protein, and wherein said vectors are packaged in said packaging cell lines.
