METHOD OF SEPARATING ALLELES

The present invention provides an improved method of separation of alleles in a sample (sample alleles), involving the use of a reference allele and further comprising the use of double stranded sample alleles and/or the use of said reference allele in a double stranded form wherein one of the strands of the double stranded alleles present has been labelled so as to allow specific digestion of one of the strands. In preferred embodiments the label is a 5'-phosphorylated group and the digestion is carried out using lambd-exonuclease. Use of this method in genotyping and/or diagnosis and kits for use in such methods of allele separation and genotyping are also included.
