PURPOSE: Plasmids for preparing DNA Ladder and a production method thereof are provided, thereby easily obtaining a large amount of DNA fragments, rapidly determining the size of sample DNA, and lowering the production costs of the DNA Ladder. CONSTITUTION: Plasmids for preparing DNA Ladder comprises pSP64 as a starting plasmid, pGDM001 having a cleavage map of figure 1, pGDM002 having a cleavage map of figure 2, and pGDM003 having a cleavage map of figure 3. The DNA Ladder composition is prepared by digesting plasmids pSP64, pGDM001, pGDM002 and pGDM003 and mixing them. A method for producing the DNA Ladder comprises digesting plasmids pSP64, pGDM001, pGDM002 and pGDM003 with EcoRI and EcoRV and mixing the digested plasmids in a specific ratio.
