An optical assembly for the detection of light from a sample, using a microscope, comprises a system to separate the illuminating light from the detection light in and/or near the plane between the eye pupil and the sample. The illuminating light is focused on the sample on the plane between it and the pupil. The optical assembly for the detection of light from a sample, using a microscope, comprises a system to separate the illuminating light from the detection light in and/or near the plane between the eye pupil and the sample. The illuminating light is focused on the sample on the plane between it and the pupil. The detection light is fluorescence, luminescence, phosphorescence, reflected light and/or diffused scattered light. The light separation system has at least one light reflection section, and at least one light transmission section, to direct the illuminating light at the sample and allow the reflected light to pass through it, or to couple the illuminating light and uncouple the detection light.
