PURPOSE: A recombinant adenovirus for expressing interleukin-18 and the gene therapy using the same are provided, thereby improving and inducing immune reaction in a cell, so that various kinds of cancer cells can be effectively treated. CONSTITUTION: Interleukin-18 protein, wherein its caspase-3 cleavage sites are substituted by an amino acid selected from the group consisting of alanine, arginine, asparagine, cysteine, glutamine, glutamic acid, glycine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, proline, serine, threonine, tryptophane, tyrosine and valine, is provided, wherein the interleukin-18 protein is matured interleukin-18 protein or interleukin-18 protein precursor; the caspase-3 cleavage site is aspartic acid of 71 or 76 amino acids in human interleukin-18 protein or aspartic acid of 69 amino acid in rat interleukin-18 protein; and the interleukin-18 protein has the amino acid sequence of SEQ ID NOs: 4, 5, 10 or 11. A gene encoding the interleukin-18 protein has the nucleotide sequence of SEQ ID NOs: 6, 7, 8 or 9. A recombinant adenovirus contains (a) a promoter regulating the expression of the interleukin-18 gene; (b) the interleukin-18 gene operably linked to a promoter; and (c) polyadenylation signal sequence.
