The invention relates to a method for carrying out the expansion of multipotent stem cells in ex vivo. The invention also relates to a two-stage method for the expansion and differentiation of multipotent stem cells in ex vivo, during which the stem cells in the first stage, i.e. during the expansion phase, can be gene-transfected. In the phase, the differentiation of the multipotent stem cells optionally ensues in cells of the hematopoietic, endothelial or mesenchymal cell line. Stem cells, progenitor cells and mature cells of the hematopoietic, endothelial and mesenchymal cell line, all of which having been obtained in the aforementioned manner, can be used, among other things, for the prophylaxis, diagnosis, and treatment of human diseases and for tissue engineering.
