PURPOSE: A method of purifying egg yolk immunoglobulins is provided which permits easy purification of egg yolk immunoglobulins from a water layer. CONSTITUTION: Egg yolk is diluted with 5 to 30 times(weight ratio) of a chitosan solution and homogenized, the pH of the solution is adjusted to a pH greater than 5.5 to cause a phase separation between an aqueous layer and a precipitate layer at -25 to 60deg.C. Thereafter, the solution is centrifuged to separate the aqueous layer from the chitosan precipitate layer. The water layer is purified with ion exchange chromatography to separate egg yolk immunoglobulins(IgY) from non-immune or immune egg yolk. The chitosan has a molecular weight of 250,000 or less.
