<P>PROBLEM TO BE SOLVED: To provide a PCR primer which causes the addition of adenine to a DNA fragment with a DNA polymerase on PCR but gives a single peak without changing a condition for the adenine addition reaction. <P>SOLUTION: The PCR is carried out by preparing four PCR primers each having an anchor sequence at the 5'-terminal and having one of A, C, G and T bases at the 5'-terminal of the anchor sequence, determining the adenine addition efficiency of each primer in each PCR, screening anchor sequences causing adenine addition to determine the anchor sequence easily causing the adenine addition, and using the primer having the determined anchor sequence. <P>COPYRIGHT: (C)2003,JPO |
