A method of amplifying a target single−stranded nucleic acid fragment with the use of a plural number of primers, characterized in that an extension reaction, wherein at least one primer is hybridized with one strand serving as a template to thereby synthesize the target single−stranded nucleic acid fragment capable of forming a loop structure and the nucleic acid fragment is employed as a template due to the loop structure, is performed while the extension reaction is regulated by a primer hybridizable with another strand which is complementary with the above−described strand. According to this method, gene examination etc. can be more easily and efficiently carried out.
