<p>The invention concerns compositions and methods for gene modification in cells, and their uses. More particularly, the invention concerns vectors comprising a) a cassette framed by sequences homologous to a target gene or by cloning sites for inserting said homologous sequences, said cassette including in 5' a bicistronic unit comprising a positive selection gene and a marker gene functionally bound by an IRES element, said genes being promoter-free, downstream of said bicistronic unit, a unit comprising a negative selection gene bound to a promoter and whereof the transcription orientation is identical or opposite to that of the bicistronic unit, and, site-specific recombination sites framing the bicistronic unit and the unit comprising the negative selection gene bound to a promoter, and b) a toxic gene located downstream of said cassette and provided with a promoter.</p> |
