Measurement method for surface measurement of chemical and biological samples using a confocal distance sensor. According to the method, individual points on the object surface are sequentially sampled. As back-scattered or reflected light quantities are small, a sensor with high numeric aperture is used and or a sensitive detector used. To determine the maximum back-scattered light intensity, different evaluation methods are used with the back- scatter signal.
