PURPOSE: A plant transformation vector pRCV1 containing a kanamycin resistance gene is provided, thereby it can be used for rescue cloning to find the function of a certain gene present in plants. CONSTITUTION: The plant transformation vector pRCV1 contains a kanamycin resistance gene, β-glucuronidase gene, an ampicillin resistance gene and bacterial replication origin. A transgenic plant Agrobacterium tumefaciens LBA4404/pRCV1(KFCC 11282) is produced with the plant transformation vector pRCV1. Primers capable of sequencing the T-border outside of the pRCV1 vector have the nucleotide sequences of SEQ ID NOs: 1 and 2. Primers capable of sequencing the T-border inside of the pRCV1 vector have the nucleotide sequences of SEQ ID NOs: 3 and 4. A method for analysis of a transformant comprises extracting genomic DNA from the transgenic plant with the pRCV1 vector, digesting the genomic DNA with Pst I and ligating the digested DNA to carry out the rescue cloning.
