PROBLEM TO BE SOLVED: To provide a new composition and method belonging to the field of molecular biology and useful for the production of DNA from a template nucleic acid and for the further DNA amplification, concretely to provide a new composition and method useful for the polymerase chain reaction (PCR). SOLUTION: The reaction liquid composition contains an anionic substance effective for promoting the DNA synthesis in an enzymatic reaction to produce DNA. Concrete examples of the anion are bivalent carboxy ions composed of dicarboxylic acid salts, especially oxalic acid ion, malonic acid ion, and maleic acid ion.
