PROBLEM TO BE SOLVED: To provide a method for producing a kit for separating and purifying nucleic acid or various biological substances, to provide a kit produced by the method, and to provide an apparatus using the kit. SOLUTION: This method for producing the kit for separating nucleic acid or a biological substance from a biological sample with a solid substance is characterized by comprising (a) a process for producing a container having chambers for holding a buffer and the solid substance for separating the nucleic acid or the biological substance, (b) a process for producing a lid which has projected portions having penetratedly formed pores and immersed in the chambers of the container and in whose chambers magnetic bars for collecting the solid substance can enter, (c) a process for charging the solid substance and the buffer into the chambers for separating the biological substance, (d) a process for sealing the container with a sealant, and (e) a process for packaging the container and the lid in cases, respectively.
