PROBLEM TO BE SOLVED: To provide a method for efficiently producing a DNA by connection of DNA fragments. SOLUTION: A fragment containing a partial sequence of a target sequence is successively inserted into a vector comprising one recognition sequence of a first restriction enzyme in which a cleavage site is located at a definite position to the recognition sequence and the cleavage site is not within the range of the recognition sequence and one recognition sequence of a second restriction enzyme in which a cleavage site is specific, capable of cleaving a fragment after being cleaved by the first restriction enzyme with the second restriction enzyme and having <=30 bases distance between the cleavage site of the first restriction enzyme and the cleavage site of the second restriction enzyme. As a result, fragments containing partial sequences are connected to each other to produce the DNA. |
