PROBLEM TO BE SOLVED: To provide a method for efficiently measuring in a short time the length of a single DNA molecule having a relatively high molecular weight of >=10,000 bp in a sample to be measured. SOLUTION: This method comprises the steps of binding the DNA in the sample to be measured to a cationic lipid added to the sample to form a polyion complex, putting the complex into extension and fixation on a solid substrate under specific conditions, and then making an image analysis of the fixed complex.
