PROBLEM TO BE SOLVED: To provide a method for producing transglutaminase, with which transglutaminase is produced by yeast and is efficiently and extracellularly released (secretory production). SOLUTION: This method for producing transglutaminase by secretory production comprises transferring an expression structure having a protransgluraminase gene sequence bonded to the downstream of a sequence encoding a yeast-derived signal peptide domain to methylotrophic yeast, culturing the methylotrophic yeast and extracellularly and efficiently releasing the obtained transglutaminase. This method for producing a large amount of transglutaminase comprises treating and cleaving the pro part of the extracellularly released protein with protease, etc. The method comprises transferring variation to the sugar chain addition part of protransglutaminase to which the yeast-derived signal peptide domain is bonded.
