PROBLEM TO BE SOLVED: To obtain a method for improving an in vitro transcription reaction, by which the efficiency of the reaction is improved. SOLUTION: This in vitro transcription reaction method for obtaining a nucleic acid transcript, comprising mixing an RNA polymerase with a template DNA having the corresponding promoter sequence, is characterized by adding tetrakis(3-aminopropyl)ammonium represented by chemical formula 1 or its salt and/or caldopentamine represented by chemical formula 2 or its salt and performing the transcription reaction at >=50 deg.C.
